检测原理
GSH-Px可以促进过氧化氢（H2O2）与还原型谷胱甘肽（GSH）反应生成H2O及氧化型谷胱甘肽（GSSG），反应式如下图。通过测定底物GSH在单位时间内被氧化的速度来表示酶的活力，反应剩余的GSH与二硫代双二硝基苯甲酸（DTNB）作用生成的黄色硫代硝基苯甲酸阴离子（TNB），测定该阴离子的浓度来计算出GSH的减少量。由于GSH在没有酶的条件下也能进行氧化（非酶反应），所以最后计算此酶活力时，必须扣除非酶反应引起的GSH减少的那部分。

样本稀释倍数
测试前先进行预实验以确定最佳取样浓度：
测血清（浆）：
将血清（浆）用PBS（0.01 M，pH 7.4）稀释成不同的浓度，取其0.1 mL按操作表进行预实验。
测组织匀浆：
测血清（浆）：
将血清（浆）用PBS（0.01 M，pH 7.4）稀释成不同的浓度，取其0.1 mL按操作表进行预实验。
测组织匀浆：
将10%组织匀浆稀释成不同的浓度，取其0.2 mL按操作表进行预实验。
测培养液：
培养液上清酶活较低，一般直接取样进行测定。
④样本最佳取样浓度探索:
本试剂盒可检测抑制率范围在20%-60 %之间，最佳抑制率范围45%-55%，其对应的取样浓度为最佳取样浓度。
注：因为酶的抑制率与活力呈抛物线关系，若抑制率大于60%时，则需将样品浓度稀释后再测试。若百分抑制率小于20%时，则需将样品浓度加大或者增加取样量。
